Protein Structure - - Bog - Oxford University Press - Plusbog.dk

Protein Structure - - Bog - Oxford University Press - Plusbog.dk

New and updated, this second edition of Protein Structure presents electrophoretic, chromatographic, and spectrophotometric techniques as well as including new chapters on the relatively sophisticated methods of mass spectrometry and ultracentrifugation. The emphasis is not on the technical aspects of the instruments, but more on the practical aspects of sample preparation and the possibilities of experimental strategy and design. The companion volume to this book, Protein Function: A Practical Approach, details comparable simple techniques for characterizing protein function. Topics in this second volume include: protein molecular weight determination by SDS-PAGE; protein analysis by mass spectrometry; immunological detection of proteins of known sequence; identification of common post-translational modifications; peptide mapping; counting integral numbers of residues by chemical modification; disulfide bonds between cysteine residues; analysis of protein conformation by gel electrophoresis; hydrodynamic properties of proteins; determining the molar absorbance coefficient of a protein; optical spectroscopy to characterize protein conformation and conformational changes; measuring the conformational stability of a protein; imunochemical analysis of protein conformation; stabilization of protein structure by solvents. Protein Structure: A Practical Approach will prove invaluable to all researchers, from PhD students to experienced group leaders.

Comprehensive Reference Work Detailing Basic and Advanced Information Surrounding Protein Engineering Protein engineering is a method of changing a protein sequence to achieve a desired result, such as a change in the substrate specificity or increased stability to the temperature, organic solvents, and/or extremes of pH or selectivity in catalytic reactions. Principles of Protein Engineering provides researchers the necessary fundamental knowledge about the protein engineering field and the rationale behind the techniques. Written by two experts with significant first-hand experience in the field, the work provides insight on protein synthesis, expression, structure elucidation, bioinformatics, directed evolution, stability, screening, and rational design. To aid in reader comprehension, the text is enhanced by short comments, feedback and personal anecdotes from pioneers and leaders in the field. Sample ideas covered in the work include: - - The fundamental knowledge of protein synthesis and expression - - Basic knowledge about alignment, homology modelling, dynamics of structure, and modeling enzyme kinetics - - The main protein engineering strategies, with a focus on directed evolution and rational design - - Differences between screening and selection, plus the main techniques for high-throughput screening Biochemists, biotechnologists, molecular biologists, and students in related programs of study can use Principles of Protein Engineering to attain both holistic and specific insight into the field of protein engineering and its numerous applications.

Protein Water Interactions - - Bog - Nova Science Publishers Inc - Plusbog.dk

Protein Water Interactions - - Bog - Nova Science Publishers Inc - Plusbog.dk

This book is aimed at understanding which molecular parameters control the thermodynamics, structure, and functions of the protein-water systems. Proteins are one of the most important classes of biological molecules. Water binding (hydration or biological water) plays a crucial role in determining the structure, stability, and functions of proteins. Knowledge of processes occurring upon hydration or dehydration of protein macromolecules is very important in biotechnological and pharmaceutical applications of proteins such as their use as biocatalysts, biosensors, and selective adsorbents. There are essential differences between hydration and bulk water surrounding a protein. This means that a characterisation of the hydration of protein macromolecules requires elucidating the effects of both the protein on water and vice versa. Therefore, a quantitative estimation of the protein and water contributions to the thermodynamic functions of binary protein-water systems is of considerable fundamental importance and practical interest. This book describes the basic principles of a novel methodology to investigate the protein-water interactions. This methodology is based on the analysis of the excess thermodynamic functions of mixing. The thermodynamic properties (volume V, enthalpy H, entropy S, heat capacity Cp, and Gibbs free energy G) of a real binary water-protein system can be expressed in terms of the excess functions. They are the difference between the thermodynamic function of mixing in a real system and the value corresponding to an ideal system at the same temperature, pressure and composition. For an ideal system, all excess functions are zero. Deviations of the excess functions from zero indicate the extent to which the studied binary system is non-ideal due to strong specific interactions between components (ie: hydrogen bonding and charge-charge interactions).

Protein Folding and Metal Ions - - Bog - Taylor & Francis Ltd - Plusbog.dk

Protein Folding and Metal Ions - - Bog - Taylor & Francis Ltd - Plusbog.dk

The role of metal ions in protein folding and structure is a critical topic to a range of scientists in numerous fields, particularly those working in structural biology and bioinorganic chemistry, those studying protein folding and disease, and those involved in the molecular and cellular aspects of metals in biological systems. Protein Folding and Metal Ions: Mechanisms, Biology and Disease presents the contributions of a cadre of international experts who offer a comprehensive exploration of this timely subject at the forefront of current research. Divided into four sections, this volume: - - Provides case study examples of protein folding and stability studies in particular systems or proteins that comprise different metal ions of co-factors - Reviews the proteins that shuttle metal ions in the cell to a particular target metalloprotein - Illustrates how metal binding can be connected to pathological protein conformations in unrelated diseases, from cancer to protein deposition disorders such as Parkinson’s disease - Addresses protein redesign of metal-containing proteins by computational methods, folding simulation studies, and work on model peptides — dissecting the relative energetic contribution of metals sites to protein folding and stability - Together, the 13 chapters in this text cogently describe the state of the science today, illuminate current challenges, propose future possibilities, and encourage further study in this area that offers much promise especially with regard to novel approaches to the treatment of some of the most challenging and tragic diseases.

Protein Purification Applications - - Bog - Oxford University Press - Plusbog.dk

Protein Purification Applications - - Bog - Oxford University Press - Plusbog.dk

Proteins are an integral part of molecular and cellular structure and function and are probably the most purified type of biological molecule. In order to elucidate the structure and function of any protein it is first necessary to purify it. Protein purification techniques have evolved over the past ten years with improvements in equipment control, automation, and separation materials, and the introduction of new techniques such as affinity membranes and expanded beds. These developments have reduced the workload involved in protein purification, but there is still a need to consider how unit operations linked together to form a purification strategy, which can be scaled up if necessary. The two Practical Approach books on protein purification have therefore been thoroughly updated and rewritten where necessary. The core of both books is the provision of detailed practical guidelines aimed particularly at laboratory scale purification. Information on scale-up considerations is given where appropriate. The books are not comprehensive but do cover the major laboratory techniques and common sources of protein. Protein Purification Techniques focuses on unit operations and analytical techniques. It starts with an overview of purification strategy and then covers initial extraction and clarification techniques. The rest of the book concentrates on different purification methods with the emphasis being on chromatography. The final chapter considers general scale-up considerations. Protein Purification Applications describes purification strategies from common sources: mammalian cell culture, microbial cell culture, milk, animal tissue, and plant tissue. It also includes chapters on purification of inclusion bodies, fusion proteins, and purification for crystallography. A purification strategy that can produce a highly pure single protein from a crude mixture of proteins, carbohydrates, lipids, and cell debris to is a work of art to be admired. These books (available individually or as a set)are designed to give the laboratory worker the information needed to undertake the challenge of designing such a strategy.
